Total RNA of sorted NCSCs was extracted using Trizol reagent and mRNA was enriched using oligo (dT) magnetic beads, approximately 1 ug of mRNA was fragmented. Then these fragments of about 200-250 bp were separated by electrophoresis. After that, cDNA fragments were synthesised using them through end repair, adenylation of the 3' ends and adapter ligation. Finally, those ranging from 250-300 bp were excised from electrophoretic gels for sequencing using an Illumina HiSeq2000 system. |
