RNA was extracted from the fruiting body of the natural-growth O. sinensis, purified using magnetic beads with oligo (dT), and then interrupted into the fragments of about 200-700 nt in fragmentation buffer, which were used as templates to synthesize the first-strand cDNA using reverse transcriptase and random hexamer-primers. The constructed cDNA libraries were constructed through A-tailing, adaptor ligation and sequenced with the Illumina HiSeq 2000 using the paired-end 125bp method. |
