XN0437T_Genome_170bp Fourteen days after germination, young leaves were harvested from HTS and HSS and used for genomic DNA isolation using a DNeasy Plant Mini Kit (Qiagen, Germany) according to the manufacturer's instructions. After quality assessment by agarose gel electrophoresis and Nanodrop (Nanodrop Technologies, Wilmington, DE, US) and quantification by a Qubit fluorometer (Thermo Fisher Scientific, Waltham, MA), the DNA was used to construct sequencing libraries by SeqCap EZ Choice Library (Roche NimbleGen, USA). Six libraries of 170 bp, 500 bp, 800 bp, 2 kb, 5 kb, and 10 kb were constructed. After the library quality was assessed by an Agilent2100 Bioanalyzer, a paired-end sequencing strategy was used on the Illumina HiSeq 2500 platform. WGS GENOMIC PCR PAIRED
