The bacterial DNA from intestinal mucosa samples was extracted using the QIAamp DNA Stool Mini Kit. Then, DNA was diluted to 1 ng/μL using sterile water and 16S rRNA genes of distinct regions (16SV4) were amplified using specific primer (515f/806r) with the barcode. PCR products from all samples were mixed in equidensity ratios and purified using the GeneJETTM Gel Extraction Kit. Sequencing libraries were generated using Ion Plus Fragment Library Kit 48 rxns. The library quality was evaluated on the Qubit@ 2.0 Fluorometer. Finally, the library was sequenced on an Ion S5TM XL platform and 400 bp/600 bp single-end reads were generated. |
