High-quality genomic DNA was randomly interrupted by ultrasound. DNA fragments ranging from 150~800 bp were recovered by electrophoresis. T4 DNA Polymerase, Klenow DNA Polymerase and T4 PNK were used to repair cohesive ends to blunt ends. The 30 end of DNA fragment was amended including an “A” base and ligated to adaptors, including a 50 end T base. All fragments were recovered by electrophoresis and then paired-end sequenced by an Illumina HiSeq 4000. |
