Total RNAs were isolated from KTRG-B48a and KTRG-B48b in 0d and 7d stages of drought treatment, and poly(A)+ RNAs were purified, and interrupted to short fragments by adding the fragmentation buffer. Next, a random hexamer-primer was used to synthesize the first-strand cDNA using these short fragments as templates, then the second-strand cDNA was synthesized using RNaseH and DNA polymerase I. Fragments were retrieved and connected with sequencing adapters. Next, fragments were selected using agarose gel electrophoresis separation, and then suitable fragments were amplified as templates. The library was paired-end -150 bp sequenced using an Illumina HiSeq 4000 |
