Total RNA was isolated by TRIZOL (Invitrogen, Carlsbad, CA), the quality and quantity of RNA were assessed with NanoDrop, Qubit and BioAnalyzer 2100 (Agilent). A RiboMinus kit (KAPA, USA) was used for ribosomal RNA depletion. Then, total RNA was divided into three equal parts after depletion of ribosomal RNA to construct a riboMinus and two riboMinus/RNase R treated replicates libraries. RNase R was purchased from Epicentre (Madison, WI) to digest linear transcripts. rRNA-depleted RNA was incubated at 37 °C with 10 U/?g RNase R in 16 ?l reaction. |
