To prepare the RiboMeth-Seq library, about 10ug of yeast total RNA was used for alkaline hydrolysis, followed by treatment of the RNA fragment with phosphatase and T4 PNK. Library preparation followed the protocol of the NEBNext Small RNA Library Prep Kit. |
