Fresh and healthy leaves were harvested and immediately frozen in liquid nitrogen, followed by storage at -80C in the laboratory prior to DNA extraction. A modified CTAB method was used to extract high-quality genomic DNA. The quantity and quality of the extracted DNA were examined using a NanoDrop D-1000 spectrophotometer (NanoDrop Technologies, Wilmington, DE) and electrophoresis on a 0.8% agarose gel, respectively. |
