Approximately 500 ml water of each sample were filtrated by filtrating membrane with 0.22 μm of aperture for collecting microorganisms. Then the filtrating membrane was cut into pieces and microbial DNA was extracted using a PowerSoil DNA isolation kit. DNA concentration and quality were checked using a NanoDrop spectrophotometer. The V4 hypervariable region of the prokaryotic 16S rRNA gene was amplified using the universal primer pair 515F and 909R. Polymerase chain reaction were performed and the amplicons were sequenced using a HiSeq system at Guangdong Meilikang Bio-Science. |
