We used a QIAGEN DNeasy Blood & Tissue kit to extract genomic DNA and sheared to fragments randomly with the Covaris system. Then the libraries were constructed through end-repair, A-tailing, adapter ligation, and purification using the NEBNext Ultra DNA Library Prep Kit for Illumina. The concentrations of the libraries were measured using Qubit2.0. The libraries were diluted to 1.5ng/ul. Then the insert size was checked using Agilent 2100 and the purity was determined using Q-PCR method. Finally, it was sequenced on Illumina HiSeq platform. |
